Eka Fibrianty – An MSc. (S2) Student of Plant Breeding and Biotechnology Major, Department of Agronomy and Horticulture, School of Graduate Study, IPB, Bogor-Indonesia, met her MSc. (S2) advisory committee for the first time on Wednesday, January 11-2012. In that meeting, Eka has presented her research plan for the MSc (S2) thesis and discussed the detail of their implementation with her supervisory committee members.

 members of her MSc. (S2) supervisory committee are Dr. Dewi Sukma (Chairman) and Prof. Sudarsono of PMB Lab., IPB (member). The title of Eka Febrianti’s MSc. (S2) thesis research will be: “Induction of protocorm-like bodies (PLB) from F2 seeds and morphological and molecular characterization of F2 populations derived from introduced Phalaenopsis F1 hybrid.”

In her research plan, she will germinate Phalaenopsis (Orchidaceae) seeds in vitro and try to generate about 200-250 plantlets. The generated plantlets will be multiplied clonally into about five seedlings. Two of them will be transferred into ex-vitro conditions and the other three will be used as explant sources for PLB induction.

For the ex-vitro activities, the regenerated Phalaenopsis plantlets will be grown in orchid’s potting media and observed for their growth, development, and other morphological characters.  Moreover, they will also be analyzed using SSR marker to determine their genetic identity. The molecular marker data will also be used to analyze the F2 of the F2 Phalaenopsis population.

For the in-vitro activities, in vitro media capable of inducing PLB from leaf of some samples of F2 Phalaenopsis seedlings will be developed. Once optimized medium for PLB induction from leaf of Phalaenopsis is identified, the developed medium will be used to induced PLB from all seedlings of Phalaenopsis F2 population (approximately: 200 seedlings). The ability of inducing PLB from leaf of the F2 Phalaenopsis individual will be evaluated.

The first advisory committee meeting run well and the advisory committee members have proposed some suggestions to improve Eka’s plan for her MSc (S2) research. Subsequently, she need to revised the MSc. (S2) thesis research proposal and prepare for the research proposal seminar. She also need to submit the revised proposal document as one of the administrative requirements for student at PBT Major. The other more important matters are to carry out the research plan and generate enough data for her thesis.

All PMB Lab staffs and associated graduate students congratulate Eka Fibrianti for finishing her first MSc. (S2) advisory committee meeting. We hope that she will be able to complete all administrative matters and generate all the necessary data for her MSc. (S2) thesis.
