As part of his PhD dissertation research, Agus Sutanto – PhD student working at the PMB Lab., Department of Agronomy and Horticulture, Faculty of Agriculture, Bogor Agricultural University (IPB) has isolated chitinase and glucanase genes from both Indonesian banana species (M. acuminata cv. Rejang and M. balbisiana cv. Klutuk Wulung). The sequences have been submitted to GenBank DNA Database.
The following are accession numbers for nucleotide sequences of the respective genes:
- JX628914: Glucanase gene (partial) from Musa balbisiana cv. Klutuk Wulung
- JX628915: Glucanase gene (partial) from Musa acuminata cv. Rejang
- JX628916: Chitinase gene (partial) from Musa acuminata cv. Rejang
- JX628917: Chitinase gene (partial) from Musa balbisiana cv. Klutuk Wulung
- JX628918: Chitinase gene (partial) from Musa acuminata x balbisiana cv. Kepok
Agus Sutanto are still isolating a couple more of chitinase and glucanase genes from an array of banana cultivars that are exhibiting different responses against Fusarium oxysporum fsp. cubense, especiallya the ‘Tropical Race 4’ strain (Foc TR4). He would like to know whether there are diversities in the level of nucleotide and amino acid sequences among those genes. Moreover, he would develop SNP markers based on nucleotide sequence diversity of both chitinase and glucanase genes and use the markers to evaluate gene diversities among Indonesian cultivars and wild type accessions of banana.
At this moment, Agus Sutanto is in his 7th semester at Plant Breeding and Biotechnology Major, Department of Agronomy, School of Graduate Program, IPB. Agus PhD program is under supervision of Prof. Sudarsono of PMB Lab. Hopefully, Agus Sutanto will be able to complete his PhD dissertation research in his 8th semester.
