[Effectiveness of poly ethylene glycol (PEG) to characterize the response of soybean genotypes against drought stress]
Polyethylene glycol (PEG) solution is able to homogeneously reduce water potential, therefore, it can be used to imitate soil water stress. The objectives of this research were to determine the effectiveness of PEG solution to characterize and evaluate the tolerance of three soybean genotypes against drought stress at vegetative growth stage. Soybean seedlings were grown on semi hydrophonic system using greenleaf as supporting medium. The seedlings were watered with half strength of MS medium for 14 days, followed by half strength MS medium containing either 0%, 5%, 10%, 15% or 20% of PEG until harvesting period.
The vegetative growth characters such as plant height, trifoliate leaf numbers, node number, leaf firing intensity, shoot dry weight, root dry weight, root and shoot dry weight ratios, and biomass dry weight were recorded. The drought sensitivity index (S) was used to group the response of tested soybean genotype against drought stress. Results of the experiment showed PEG solution inhibited vegetative growth of soybean plants, as indicated by reducing plant height, nodes, leaf number, and biomass dry weight. Moreover, the stress due to PEG solution resulted in increasing leaf firing intensity, root length and root/shoot dry weight ratio. The drought sensitivity index calculated based on the collected data have grouped the three tested soybean genotypes into drought tolerance (MLG2805), medium tolerance (MLG2999) and drought sensitive (MSC8606). This drought stress response was similar to results of the previous evaluation under field condition. Under this condition, PEG solution at 5% concentration was the best for differentiating the response of soybean genotypes against drought stress.
